Tre anni dopo essere finito dietro le sbarre, il rapinatore armato Jake Blues viene liberato per buona condotta. Ad attenderlo fuori dal carcere c'è il fratello Elwood, una ex fidanzata inferocita, la notizia che l'orfanotrofio dove sono cresciuti sta per essere chiuso e l'illuminazione divina. Convinti di essere in missione per conto di Dio i Blues brothers riuniscono con le buone e le cattive la vecchia band e organizzano un grande concerto benefico mentre fuggono da polizia, una banda di musicisti, il proprietario di un locale e i nazisti dell'Illinois che li vogliono fare fuori.
